Xenical (Generic Orlistat) is a specific inhibitor of gastrointestinal lipases.
As a result, the enzyme that has lost activity is not able to break down the fats that come from food in the form of triglycerides.
Under the work of the drug, 24–48 hours after its administration, the concentration of fat in the stool mass increases. By reducing the fat depot, the tool provides control of body weight. For the effective action of the drug does not require its absorption into the systemic circulation. When taking Xenical 3 times per day by 120 mg, then 30% absorption of fat from food is blocked
Systemic exposure of the substance is minimal. The maximum radioactivity in plasma was observed after about 8 hours. The content of unchanged orlistat was close to the limit of detection (less than 5 ng/ml).
In therapeutic studies that included monitoring plasma samples of patients, unchanged orlistat was detected irregularly in plasma and at low concentrations - less than 10 ng/ml. Signs of cumulating of the drug is not detected, which confirms its minimum absorption. In vitro, the active substance binds to plasma proteins by more than 99%, mainly with albumin and lipoproteins. In the red blood cells, Xenical is able to penetrate in minimal quantities.
Indications for the treatment is, first of all, obesity, including the provision of reducing and maintaining body weight, in combination with a low-calorie diet. This medicine reduces the threat of re-gaining body weight after achieving its initial reduction.
